The tissue culture technique of Zanthoxylum nitidum( Roxb. ) DC.
Abstract
The tissue culture technology of two types of Zanthoxylum nitidum( Roxb.) DC. was studied in this paper. The results showed that when stem segments and apical buds were used as explants,the adventitious buds initiating medium was MS + 6-BA 0. 4 mg / L + IBA 0. 2 mg / L. The best proliferation mediums were modified H + 6-BA 0. 6 mg / L + NAA 0. 3mg /L and modified MS + 6-BA 0. 6 mg / L + IBA 0. 3 mg / L,and the proliferation coefficients were 4. 5 and 4. 1,respectively. The optimal rooting culture mediums were 1 /10 MS + NAA 0. 5 mg / L and 1 /2 MS + ABT60. 6 mg / L + IBA 0. 4 mg /L,and the rooting rates were 96. 2% and 90. 2%,respectively,the transplanting survival rates were 95. 2% and 88. 9%respectively on yellow subsoil,fine sand and peat soil at the rate of 3∶ 1∶ 1. The difference of morphology between two types of tissue culture seedlings was significant.
