A Preliminary Study on Tissue Culture Technology of Camellia oleifera
Abstract
Seed embryo or 2-year-old cuttings sprout shooting of Camellia oleifera were used as explants for tissue culture propagation technology research. The results were as follows: the suitable induced medium of C. oleifera seed embryo was MS + 6-BA 2. 0 mg /L + NAA 0. 2 mg /L + GA31. 0 mg /L + sucrose 30 g /L + carrageenan6. 5 g /L,and its suitable proliferation medium was MS + 6-BA 1. 5 mg /L + NAA 0. 1 mg /L + GA31. 0 mg /L + sucrose 30 g /L + carrageenan 6. 5 g /L,the multiplication rate was 3. 50. The suitable induced medium of axillary bud was MS + 6-BA 1. 5 mg /L + IAA 2. 0 mg /L + GA31. 0 mg /L + sucrose 30 g /L + carrageenan 6. 5 g /L,and its suitable proliferation medium was MS + 6-BA 2. 0 mg /L + IAA 1. 0 mg /L + GA31. 0 mg /L + sucrose 30 g /L + carrageenan 6. 5 g /L,the multiplication rate was 3. 96. The fine sand was a suitable matrixes for rooting of highly lignification shoots,the rooting rate was 95%.
