Abstract

This repository contains experimental protocols developed and used within the PRIN project “RiWeCIS”, funded by the Italian Ministry of University and Research (MUR). Due to Zenodo platform limitations, the repository is provided as a compressed (.zip) archive in order to preserve the original folder structure. Users are required to extract (unzip) the archive to properly access and navigate its contents. The repository includes protocols associated with different Work Packages (WP2 and WP3), detailing experimental procedures applied during the project. ----------------------------------------WP2 Protocols---------------------------------------- The repository contains the following protocols: - Plant growth protocol Description of the procedures for seed sowing and growth conditions of the plant species involved in the project. - VIGS protocol using TRV A detailed protocol describing Virus-Induced Gene Silencing (VIGS) using Tobacco Rattle Virus (TRV), including all steps from cloning to plant treatment, applied to *Solanum nigrum* and *Nicotiana benthamiana*. - VIGS protocol in *Echinochloa crus-galli* Protocol describing attempts to establish VIGS in *Echinochloa crus-galli* using: - Cucumber mosaic virus (CMV) - Barley stripe mosaic virus (BSMV) ----------------------------------------WP3 Protocol---------------------------------------- - Greenhouse treatment with nano-encapsulated herbicide This protocol describes greenhouse experiments conducted on *Oryza sativa* and *Echinochloa crus-galli*, aimed at evaluating the efficacy of nano-encapsulated imazamox. The protocol includes details on plant growth conditions, preparation and application of the nanoparticle-based formulation, treatment setup, and assessment of herbicide effects at the whole-plant level. ---------------------------------------- All protocols are provided to ensure reproducibility and transparency of the experimental procedures developed within the PRIN project “RiWeCIS”.
