Objective: Comparative study clinical and vasoprotective efficiency of 12-weekly monotherapy with lercanidipine and its combination with perindopril in hypertensive patients. Design and Methods: There have been included 60 hypertensive men with the I-II grade hypertension. Middle age was 50.6 ± 7.1yrs. and average duration of hypertension 6.5 ± 5.1 yrs. Blood pressure was measured with sphygmomanometer by Korotkov method. Flow-mediated endothelium dependent vasodilatation (EDVD) was measured during reactive hyperemia due to 5 minute brachial occlusion. Patients were divided into 2 groups according to therapy: monotherapy group included 40 patients and the combination group included 20 patients. Daily dose of lercanidipine was 10–20 mg and the perindopril – 5–10 mg. Results: It has been shown high antihypertensive efficacy of lercanidipine monotherapy and its combination with perindopril along with the absence of any side effects. SBP reduction was -15.8% in the monotherapy group: from 151.2 ± 10.1mmHg to 127.4 ± 10.9mmHg (p = 0.000) and -16.1% in the combined group: from 152.6 ± 8.9mmHg to 127.9 ± 7.3mmHg (p = 0.000). DBP has decreased -14.8% in the monotherapy group: from 98.0 ± 5.8mmHg to 83.4 ± 7.7mmHg (p = 0.000) and -15.7% in the second one: from 96.8 ± 6.3mmHg to 81.3 ± 3.6mmHg (p = 0.000). 84.2% of patients who took combined therapy have archived goal BP (<140/90mmHg) in comparative to those who were in monotherapy – 62.5%. Moreover, it has been noted, significantly improvement of EDVD during both of therapy mode. δD has changed from 3.7 ± 3.1% to 8.7 ± 4.8% in the monotherapy group (p = 0.000) in contrast with second group: from 5.5 ± 3.8% to 10.3 ± 3.8% (p = 0.000). Conclusion: Results of our study have shown high antihypertensive efficiency of lercanidipine. Besides, its combination with perindopril has promoted to full normalizing of EDVD.
