Usporedba ELISA i LC-MS/MS tehnike u analizi aflatoksina u kukuruzu

Аннотация

Aflatoxins are toxic secondary metabolites of Aspergillus spp. The aim of this study was to compare the positive results of aflatoxins obtained by ELISA assay with the confirmatory LC-MS/MS method. Analysis was performed on maize, harvested in 2012 which was characterized by very hot and dry weather, and the samples were collected in 2013 in eastern Slavonia. The LC-MS/MS method was validated with recovery range from 79 to 96 %, detection limit from 0.5 to 1 μg kg-1 and limit quantification from 1.65 to 33 μg kg-1. The LC-MS/MS results showed the following aflatoxin B1:B2:G1:G2 ratio: 1:0.08:0.18:0.02. The highest value obtained by LC–MS/MS refers to 233.57 μg kg-1 aflatoxin B1, while the highest aflatoxin B1 value obtained by ELISA was 1710.03 μg kg-1. The results obtained by ELISA were higher for 364 ug kg-1 on average compared to the LC-MS/MS results. Taken together, the results point to an advantage of ELISA assay due to its applicability for rapid screening and detection of aflatoxin presence. However, the results have to be confirmed using a more precise technique, which in this case was the LC-MS/MS method.
