Studies on the Active Principles from Aconitum flavum Hand-Mazz. The Structures of Five New Deterpenoid Alkaloids

Reinvestigation of Aconitum f l a w Hand-Maze r e s u l t e d i n t h e i s o l a t i o n of f i v e new diterpenoid a l k a l o i d s , i .e . .dehydronapelline (1 ).12-acetyllucidusouline ( 2 ) .1-epi-napelline ( 3 ) .12-epi-napelline (41, and I-demethylhypaconitine ( 9 ) .along with n a p e l l i n e , lucidusculine.aconitine.3-acetylaconitine, deoxyaconitine, flavaoonitine, beneoylaconine.and neoline.The s t r u c t u r e s of these new compounds were determined based on spectral data and chemical transformations.The pharmacological t e s t s showed t h a t 12-acetyllucidusculine ( 2 ) has a s t m n g e f f e c t of lowering of heart r a t e i n Langendorff's guinea pig h e a r t s .corresponding t o C22H33N03 (M' 359.2466, calc.959.2460) by high r e s o l u t i o n ms.The i n f r a r e d spectrum showed absorptions at 3400 (hydroxyl).1660 and 890 om-' (exomethylene).The mass spectrum of t h i s base w a s In acoord with t h a t of napelline (5).Every carbon chemical s h i f t w a s very s i m i l a r t o corresponding value of napelline ( 5 ) =",'LI-P 'be" rX9',,-" .be"'09.Z '91'1 'G5.I ' Z C ' I *LL'5 ' ' 8 t .5 '16'.'
