Objective: To study the role of carrying G460T polymorphism of Alpha-Adducine gene in development of left ventricular hypertrophy in Uzbek hypertensive patients. Design and method: The study included 78 male and female Uzbek patients with stage I-II AH (ESH/ESC 2013) with an average age of 49.54 ± 10.20 years. Mean duration of arterial hypertension was 5.77 ± 4.11 years. M- and B-mode echocardiographic examination was performed. Left ventricular mass index (LVMI) was calculated as a relation of left ventricular myocardium mass to body surface area. Presence of left ventricular hypertrophy (LVH) was diagnosed when LVMI>115 g/m2 in male and >95 g/m2 in female. Genomic DNA extracted from peripheral blood. PCR was performed. Analysis of variance (ANOVA) was done using statistical software packages in NCSS 2003, PASS 2002 (Hintze, 2001) and SPSS 11.5 (Levesque, 2007). In all tests significance threshold value was set at p < 0.05. Results: After genotyping patients were divided in to three groups: carrier of TT genotype – 2 patients (2.6%), carrier of GT genotype – 33 patients (42.3%) and carrier of GG genotype – 43 patients (55.1%). A significant differences in structural parameters of left ventricular were found in patients depending on genotype. Thus, the interventricular septal thickness was 12.6 ± 0.08 mm in patients with TT genotype versus 10.5 ± 0.12 mm in patients with GT genotype and 10.9 ± 0.17 mm in patients with GG genotype (p < 0.05); the posterior wall thickness was 12.8 ± 0.25 mm, 9.2 ± 0.14 mm and 9.5 ± 0.16 mm respectively (p < 0.05). The significant trend to high value of left ventricular mass (LVM) was found in account with G460T polymorphism of alpha-adducin gene. LVM was following: 361.95 ± 77.84 g in patients with TT genotype versus 256.18 ± 63.28 g in patients with GT genotype and 263.59 ± 75.40 g in patients with GG genotype (p > 0.05). The LVMI also was had the trend to high value in patients with TT genotype: 163.48 ± 29.92 g/m2 versus 132.45 ± 35.56 g/m2 in patients with GT genotype and 127.55 ± 27.61 g/m2 in patients with GG genotype (p > 0.05). Conclusions: Thus, the carriage of TT-genotype of G460T polymorphism of alpha-adducin gene was associated with severity of LVH degree in Uzbek hypertensive patients.
