Influence of all-trans-retinoic acid on cell shape and apoptosis of thyroid cancer cell lines

OBJECTIVE:To observe the impact of all-trans-retinoic acid(ATRA) on cell morphology and apoptosis of thyroid cancer cell lines (FTC-133, K1, 8505C) in vitro. METHODS: Treated with different dose of ATRA(final concentration of 0,10 -7 ,10 -6 ,10 -5 and 10 -4 mol/L), the cells were observed with a convert-microscope in shape and with a fluorescence-microscope in apoptosis dyed by Annexin-V FITC/P.RESULTS: Compared to the controls, the cells of FTC-133 grew shuttle-likely and arranged more regularly and gathering of K1 cells decreased. No shape change was found in 8505C cell line, but there was a reduction in the ability of adhering to wall at 10 -4 mol/L ATRA. Apoptosis rates were (7.8±2.6)% and (11.6±3.7)% at high dose of ATRA (10 -5 , 10 -4 mol/L) in FTC-133 , and (7.9±2.6)% and (10.8±3.3)% in K1, respectively. It was (15.3±4.7)% at 10 -4 mol/L of ATRA in 8505C. It was significantly higher compared with that of the control group[(2.8±0.7)%,(3.1±0.9)%,(6.7±2.2)%, P0.05].CONCLUSION: ATRA can induce morphological changes and apoptosis in these three thyroid cancer cell lines.
